Di-tert-butyl dicarbonate application:
1. Amino group protection (Boc protecting group)
Used to protect the α-amino group of amino acids (such as peptide synthesis) to prevent side reactions.
Protect primary and secondary amines in organic synthesis (such as drug intermediate synthesis).
The Boc group can be selectively removed under acidic conditions (such as TFA/hydrochloric acid) and has good stability.
2. Medicine and peptide synthesis
Used to synthesize active molecules such as antibiotics, anti-tumor drugs, and enzyme inhibitors.
Widely used in peptide solid phase synthesis (SPPS), such as the synthesis of bioactive peptides such as insulin and hormones.
3. Organic synthesis intermediates
Used to construct tert-butyl carbamate (Boc-amine), as a key intermediate to participate in coupling reactions (such as Buchwald-Hartwig reaction).
Widely used in the total synthesis of heterocyclic compounds and natural products.
4. Biochemical research
Modify the amino group of proteins or enzymes for structure-function studies.
Protect specific amino acids in chemical protein synthesis (such as native chemical ligation, NCL).
